Versatility of the Nudix Enzymes
Nudix hydrolases are a superfamily of pyrophospho-hydrolases catalyzing the hydrolysis of a nucleoside diphosphate linked to different moieties. Some of the substrates are metabolites that require modulation and the enzymes control those levels. We have defined the structural basis of the versatility of Nudix enzymes through the structural determination of archetype enzymes of the different families such as the ADPRases, CoAses, NDPases, CDPChases and UDPGases. Our work aims to elucidate rules for identifying families according to their catalytic activity, tertiary structure, quaternary arrangement and phenotype.
